Waynesboro was founded in 1799 by Captain John Waynes and his men as a post on the Susquehanna River. The town grew rapidly, and in 1825 it became the seat of government for what is now Lancaster County. In 1864, Waynesboro became the county seat of Lancaster County. By 1870, the population had grown to over 10,000 people. The town's growth led to its first school, erected in 1814, being replaced in 1868 by a larger building that still stands today. In 1865, Waynesboro became part of the newly created Commonwealth of Pennsylvania. In 1971, it rejoined the United States as part of the new country of America. In 1992, the town annexed parts of neighboring Westmoreland Township. Today, Waynesboro is an agricultural center with a large population of migrant workers from around the world who participate in its agriculture industry. It also has a number of businesses that provide services such as retail stores and restaurants.

It is part of the Pittsburgh metropolitan area. The town was founded in 1768 by George Washington and his family as part of the Allegheny Valley district. It was named for Major General John Wayne, commander of American forces during the French and Indian War (1763-1765). After the war, Washington returned to America and settled here with his family. In 1801, he built a house on what is now Main Street. He served as justice of the peace here from 1802 until his death in 1827. His wife Lucy also lived here for some time before moving to Mount Lebanon, Pennsylvania. Waynesboro had a sawmill and gristmill on its main street until 1912 when they switched to mechanical operations under license from Girdwood Mill Company in nearby Monongalia County. The mill closed in 1944 but has been restored several times since then as an openair museum with exhibits on local history and industry. The first post office here opened in 1800; it moved to its present location two years later. In addition to its rich heritage including two Presidents (George Washington and James K Polk) who were born nearby, Waynesboro also has a number of notable individuals associated with it: including actor/director William Saroyan, educator James Atherton (founder of West View Elementary School), writer Ayn Rand (!), industrial designer Harry Blackman (founder of Air Products & Chemicals), singer Bobby Vinton (the first human being ever documented singing publicly while free), politician Scranton Mayor Tom Foley (!), entrepreneur Milton Bradley (!). It was ranked "Best Place To Live" by Forbes magazine twice between 2006-2011 - once for quality of life issues such as low crime rates and accessible public transportation options, and again for having "a sense that it's home". Its residents have even created their own zip code - 71801 - which may soon become obsolete given that neighboring municipalities have all annexed into Pittsburgh since 2010!

It is part of the Allentown-Bethlehem-Easton, PA Metropolitan Statistical Area. The first Europeans to arrive in Waynesboro were Dutch settlers in 1685 who founded a trading post on the corner of West Market and South First Streets. The area now known as Waynesboro was first settled by English colonists in 1724 when they arrived from Northampton County, England on their way to settle Philadelphia. George Washington and his men traveled through Waynesboro on their way to battle against the British at Yorktown in 1781. In 1811, General James Burnside and his brigade invaded Pennsylvania during the Mexican War and burned many villages and towns along his route before reaching Waynesboro. In 1814, John Dickinson appointed Colonel Isaac McCoy as commander of Militia forces stationed at present day Easton for defense against Indian incursions into western Pennsylvania. McCoy's troops fought at Battle of Gettysburg while supplies were brought through Waynesboro for that engagement. In 1865, Confederate General Robert Ewell's army attacked Union Brigadier general Edward Pemberton's force near Reading Pike in what became known as Battle of Round Top; Pemberton's force was destroyed with barely any loss to Confederate units while General Lee withdrew across Monmouth Creek without engaging Pemberton himself. Allentown Businessman Abraham Lincoln was born here on November 6th,1809; he later served two terms as President Ulysses S Grant’s Vice Presidential running mate after losing an election that year to Andrew Johnson via stolen ballots. After leaving office Lincoln moved back to Illinois until he died here January 15th, 1865 seven months after being shot by John Wilkes Booth while attending a campaign rally near Ford’s Theatre in Baltimore Maryland
