The Gloucester Artillery Institution was founded in 1834 by Colonel John Brown, a veteran of the American Revolution. The institution became an army reserve artillery regiment in 1847 and then an artillery brigade in 1861. In 1865 it was redesignated as the 1st Massachusetts Infantry Regiment. The regiment served in the Union Army during the Civil War and surrendered at Appomattox Court House on April 9, 1865. The regiment's colors were retired in 1898 and remain display at Armory Square in Gloucester. The 1st Massachusetts Infantry Regiment is now part of the 4th Brigade, 3rd Division, Massachusetts National Guard.

In 1853, Charles Fenton established an insurance business in downtown Gloucester which soon became one of the largest insurance companies in New England and later expanded into other businesses including real estate brokerage and banking before folding in 1966 and becoming part of Travelers Insurance Companies Incorporated.. In 1865, Abraham Lincoln's great-grandson George Washingtons moved from Springfield to Gloucester with his family for farming purposes; he started a grocery store here that became one of the most successful small businesses in town throughout its history in 1893, Julius Caesar Weinshall opened what is now known as Weinshall's Market across from Union Station (now Macy's) - it would be renamed Sams Club after Sam Adams Brewing Company bought it in 1978 in 1911, Averill Harriman began trading at 7 Main Street - it would become Harriman's Furniture Mart before being acquired by Sears Roebuck & Co., headquartered there until 1993 in 1915, First National Bank merged with Trustco Bank to form First Trust Corporation which became Prudential Financial Corporation operationally within Prudential until its merger with still another trust company called Donaldson Lufkin & Jenrette LLP (DL&J) back into Prudential Incorporated on November 3rd 2006; DL&J remains based at 7 Main Street today under their new name Jardine Matheson LLP
